This is really interesting -- thanks for sharing. It seems to me that you could probably have nginx running on a regular box and then CloudFront as a caching CDN to avoid the S3 update delay.

I'm curious to see what you've done -- anything you can share?. I'll let John write up the details in a future blog post, but mostly there are a lot of strange edge cases and problems that have to deal with maintaining closures, etc.
