I was once very skeptical of the amyloid hypothesis, but I think the genetic evidence is very convincing. Most (all?) cases of familial early onset AD are caused by mutations that increase amyloid. And there is a rare variant of the amyloid gene that is completely protective against AD. Plus the efficacy of Leqembi.
