Yes, if you make the vehicleLocations request without specifying a route (http://webservices.nextbus.com/service/publicXMLFeed?command=vehicleLocations&a=sf-muni&t=0 http://webservices.nextbus.com/service/publicXMLFeed?command...) you get the locations of all the vehicles at once.
