It's been done. Singapore Airlines flew the longest scheduled flight (between New York and Singapore, SQ21 and SQ22) using an all-business A340-500 [1]. They canceled the service in 2013 since revenue wasn't high enough. Apparently, there are a few small all-business carriers in operation [2].
