Ask some questions like what kind of development processes they follow, how they organize themselves and what kind of offsite activities they have. You can tell a lot from the answers about the culture in general. For example do they respect employee's time and do they show good team collaboration and cohesion or are they a 'hero/special snowflake culture'. If you don't like the answers, then it might not be a good fit for you.
